Glyoxylic-oxime C-N coupling (oxalic acid + nitrate to glycine)

Glyoxylic-oxime C–N 커플링 (옥살산 + 질산염 → 글리신)

옥살산과 질산염으로부터 글리신을 합성하는 Cu-Hg 촉매 3단계 경로 (paper_181, see Kim et al., Angew. Chem. Int. Ed. (2021)). C–N 결합은 glyoxylic oxime 중간체를 통해 형성되며 — glyoxylic imine이 아님 — 이는 고전적인 카르보닐 + 아민 환원적 아민화 경로와 구별된다.

단계

  1. 옥살산의 2e⁻ 환원 → 글리옥실산 (개시 전위 −1.0 V vs Ag/AgCl; oxalic-acid-CO₂-derived).
  2. NO₃⁻의 6e⁻ 환원 → 하이드록실아민 (NH₂OH) (개시 전위 −1.1 V; 선택적 NH₂OH 생성 F.E. 66.8% 별도 확인, Fig. S₈).
  3. NH₂OH의 글리옥실산 친전자성 알데하이드 탄소에 대한 친핵성 공격 → glyoxylic oxime + H₂O (화학적 C–N 결합 형성; oxime 개시 전위 −0.7 V vs Ag/AgCl).
  4. Glyoxylic oxime의 4e⁻ 환원 → 글리신 (glycine-electrosynthesis-product).

총 화학량론적으로 글리신 1분자당 12개의 전자를 소비한다 (see Kim et al., Angew. Chem. Int. Ed. (2021), Fig. 5c; Eqs. 1–5, p. 22119).

증거 및 선택성

  • ¹H NMR을 통해 전해된 용액에서 글리옥실산, 글리콜산, 암모니아, 및 glyoxylic oxime이 검출되었다 (Fig. 4e).
  • 대조 실험 결과: 글리옥실산 + NH₂OH → 글리신 생성, 글리옥실산 + NH₃ → 글리신 미생성 — 이는 oxime 경로(imine 아님)를 확정한다.
  • 선택성은 전극 표면 피복률이 아닌 벌크 중간체 농도(글리옥실산과 NH₂OH는 Cu-Hg 표면에서 쉽게 탈착됨)에 의해 결정된다. 누적 전하량이 400 → 4000 C으로 증가함에 따라 글리옥실산 농도는 0.022 → 0.046 M로 상승하고, 글리신 F.E.는 26.1% → 43.1%로 향상된다 (Fig. 5d).
  • 최적 옥살산 : NaNO₃ 비율 = −1.4 V에서 1:1 (2:1 또는 1:3의 경우 F.E.가 각각 22.1% / 18.5%로 감소; Fig. 5b).
